Dancevic, Vasek Pospisil and Aleksandra Wozniak join Rebecca Marino, who didn't need to go through the qualification route.
Dancevic got past Bradley Klahn of the U.S. 3-6, 6-1, 6-3.
It's the first time the 26-year-old from Niagara Falls, Ont., has made the men's draw at the tournament since 2008. He's been dogged by injuries since then.
Dancevic's opening round match will be against Marsel Ilhan of Turkey, who's currently ranked 100th. The winner will then face either No. 19 Fernando Verdasco of Spain or Finnish vet Jarkko Nieminen.
Vancouver's Pospisil defeated France's Charles-Antoine Brezac and Slovenia's Grega Zemlja to get into the main draw.
Pospisil was a 6-1, 7-5 victor over Brezac, and outlasted Zemlja 6-4, 2-6, 7-6 (5).
Pospisil will debut next week against No. 65 Lukas Rosol of the Czech Republic. The survivor of that match could then draw No. 25 Feliciano Lopez of Spain in the second round
Wozniak downed Switzerland's Stefanie Voegele and Israel's Julie Glushko to qualify in the women's singles draw.
The Blainville, Que., native won 6-4, 5-7, 6-2 over Voegele and 6-1, 6-4 over Glushko.
Wozniak will face Christina McHale to begin her draw. The American teen has split two matches with world No. 1 Caroline Wozniacki this month.
The Wozniak-McHale winner could be in line for a second round date with former Wimbledon finalist Marion Bartoli of France, the tournament's No. 8 seed.
Stephanie Dubois of Laval, Que., fell 7-6 (1), 3-6, 6-1 to Germany's Tatjana Malek in qualifying.
Dubois will be first in line for a so-called "lucky loser" spot should one become available through a pullout.
Vancouver's Marino, ranked 39th in the world, had a memorable first U.S. Open in 2010. She won her first round match and then gave seven-time Grand Slam champ Venus Williams a tough test before bowing out.
Marino didn't get an easy first round assignment in Thursday's random draw. She's pitted against Argentina's Gisela Dulko, who once reached No. 26 in the WTA rankings. Dulko is currently ranked 58th in the world.
No. 4 Victoria Azarenka of Belarus could loom in the second round for the winner.
Canada's top-ranked men's player will miss the tournament. Milos Raonic of Thornhill, Ont., is still recovering from a hip injury suffered at Wimbledon.
It remains to be seen if the matches will begin as scheduled in Queens on Monday, with Hurricane Irene due to threaten the area, according to the weather forecast.
The annual Arthur Ashe kids' day, scheduled for Saturday, was cancelled due to expected poor weather.
