A non-import who grew up in Pickering, Ont., he was named the league's most outstanding offensive lineman in 2003.
"Andrew Greene brings a tenacious attitude and a wealth of experience," Blue Bombers general manager Brendan Taman said in a statement.
Greene played 116 games for the Roughriders before being released Feb. 1.
The Indiana product also enjoyed stints with the Miami Dolphins, Seattle Seahawks and Jacksonville Jaguars.
"His addition will make an already solid unit that much stronger this coming season," Taman said.
Also Wednesday, Corey Chamblin was named Winnipeg's defensive backs coach.
Chamblin, 27, has NFL experience with the Baltimore Ravens, Jaguars, Green Bay Packers, Tampa Bay Buccaneers, Denver Broncos and Indianapolis Colts.
He also played briefly for the Rhein Fire of NFL Europe and spent last season as an assistant coach for the Frankfurt Galaxy.
"I became very interested in Corey when I learned how varied his background was as a player and as a coach," Blue Bombers head coach Doug Berry said. "Obviously, we would like to have someone who will continue the enhancement of teaching the DBs we have here.
"I felt that Corey, with all the experience he has, would fit that bill."
